Lehmanns is Berlin's largest academic bookstore, and you'll find an excellent selection of specialist literature in English here. The computer / IT section is especially good.
There also is a small selection of Berlin-related books, some in English, on the ground floor. The gallery between the ground and first floor contains a decent selection of travel guides. On the third floor there's a small café.
Note: the building used to be occupied by Kieperts, which was Berlin's largest general book shop, but which went bankrupt in 2002 following a misguided attempt to become a trendy multimedia store.
The building which Lehmanns occupies, the Hardenberghaus, is a particularly well-regarded example of 1950's architecture.
